Business upgrade at checkin is not possible, unless you change your booking class.
Premium economy upgrade is usually possible if they have seats available. Vary from $100 to $300.
I've tried the premium economy and, besides the seat and champagne, there is no big difference from regular economy.
That said, premium economy seats are VERY uncomfortable and I get numb legs all the time.
Personal opinion, not worth it.
