From my understanding, it's best to put it closest to the entry point. We have cat-5 cable in almost every room, so it would be possible to put it on the first floor in one of the rooms.
I would test it on the first floor. If it doesn't make a difference you could always put it back where it is. I have not had good experience with extenders. If I could figure out how to make them work as advertised, I would be so happy.
