The LAX to BUF flight will be with an all coach seat configuration. Mint is only offered from LAX/SFO to JFK. Boston will be added soon and there are a couple of Caribbean routes from JFK and Boston that have or will have Mint.
The Mint product is excellent and often offered at a very reasonable price. You can google JetBlue mint to view the many blogs and articles that have written about it.

TrueBlue is pretty straight forward. It is not a program with a lot of tricks to figure out. You earn 3 points per dollar for flights plus another 3 points if you book on jetBlue.com. Redemption is also based upon the price of the flight in dollars so the amount of points required gets cheaper when the fare goes down and more expensive as the price increases. Redemptions are always available on every flight that is not sold out as long as you have enough points.
Mosaic status can be earned either by taking 30 flights and earning 12,000 status points (basically buying $4,000 worth of tickets) or earning 15,000 status points with no requirement to fly a particular number of flights. You earn 3 status points for every dollar spent on JetBlue flights.
A couple times of year, they usually offer double status points. You have to register for those. One is ongoing through February. The other time is usually towards the end of the year.
