OP, your first quote deals with what counts towards spend. Your donation will count towards your annual spend. I tithe to my church via AMEX payments. It has always counted towards my spend, as have donations I've made online to other organizations.

The second quote only deals with the discount you receive for onboard purchases (such as food/drink, etc.) The second quote is saying that you won't get a discount for charity payments onboard the flight. The only thing I've ever seen them sell that would probably apply to that would be the special drinks they serve on cancer awareness month. (but there may be more). In this case you wouldn't get a discount on that special drink, but you would still get spend credit.
