Have been making charitable donations for years through AMEX and it has always counted towards spend. This includes auction items from charities. AMEX isn't analyzing what the charge is for; it could be a lot of things.

My one warning would be is to make sure that the contribution is actually "charged" by the charity during 2015 or it won't count towards this year. They usually want to get their money right away, but if there is a problem, you could have a problem.
