I don't think that this is a planned A380 route in the near future at all, but you can fly the A380 to JNB and then connect on Comair if you want. Just be aware that there is no airside connection and it can add quite a bit of flight time.
We are strongly considering this option as its also slightly cheaper.

I have looked at going via the middle east but it's close to an extra 6 hours of travel and travelling with the kids was want to avoid. Also, we can't afford to fly in anything above economy as its a family vacation.
