I have stayed at the DIA and to answer your questions:

1. When you land and I am assuming you are in transit, there will be a hotel rep waiting at the Arrival gate. They will take you to Security check because you need to access the hotel from the departure/duty free level.

2. There are smoking rooms available at DIA, so just ask for one.
