ANY expense (not just travel) counts toward your sign up bonus spend requirement.
Any qualifying travel expense can be reimbursed using whatever points balance you have when you try to redeem them.
They're all just points and travel expenses. Nothing special or unique about either. Once a qualifying ($25 or new $100 min) travel expense has posted, any points balance at that moment in time or later can be applied (subject to $25 increment if less points balance than expense).
