Can you redeem miles on a purchase (hotel) on a statement that you hit the spend?

For example - I have a $500 hotel stay on 9/1/15 and my statement closes on 9/15. That $500 stay helped me hit the spend bonus. Can I use the 40k bonus to pay for part of the $500 hotel stay on 9/1?
