Booking itself does not cost you any miles until you ticket (any segment). However, I did automatic ticketing before (when seat was ready, the upgrade was ticketed).

Whether your waitlist can be cleared depends a lot on your segment, the cabin, the date, and your Marco Polo status. If you must travel, I would recommend getting a revenue ticket instead.
