As a base for travelling, I think it's location next to Green Park station is hard to beat. While I don't really like the atmosphere, there isn't much to complain about the hotel.
Leicester Square is rather loud and full of drunk bridge and tunnel types which makes the Hampshire a bit unappealing.
While not disagreeing with anything (apart from the tone of the 'bridge and tunnel' reference), to put things into perspective for the OP - it's a 5 minute walk from Mercer Street to Leicester Square.
