Each time you go to Zones 3-6, you will have between £1.50 and £2.80 deducted from your credit balance (it's £1.50 off-peak, at peak time it will vary depending on where you go).

You can take the Underground direct from Heathrow to Caledonian Road, the hotel is about a 15 minute walk from there (This will cost £1.50 or £2.80, as above).
The underground journey will take about an hour.
