AA only operates JFK-LAS once daily, departing JFK in the late afternoon, generally timed for connecting pax coming from the mid-day arriving European flights.
Like anything with LAS, timing is everything. If you're flying in or out around a big convention or other event, upgrades can be tough to nonexistent even for EXPs. But on a random Tues or Wed in the winter, you should have a decent chance.
