Yes. Swipe the same card twice on the bus, and when you swipe that card at the subway it will know that you've got two transfers and let two people pass through the turnstile on a single swipe.
Correct. If you swipe the card twice at the subway turnstile, an additional $2.50 will be removed from the card's stored value.

And the same thing works in reverse: If you start your trip on the subway, swipe the card twice at the same turnstile before proceeding through; when later transferring to a bus, dip the Metrocard just once into the farebox. The farebox should emit two beeps, and the display should read "2 Transfers" (or something similar).

When boarding a bus with two or more free transfers stored on a Metrocard, I usually alert the driver just before I dip the card, so that (s)he is alert to the fact the multiple transfers are about to be used.
