Each flight is charged separately so if you flew from New York to Madrid to Barcelona you'd be charged the miles required for New York to Madrid and separately additional miles for Madrid to Barcelona. It doesn't matter whether you connect straight through or have a stopover in Madrid.
Open jaws can most easily be done by booking 2 one-ways.

Do I understand correctly that open jaw doesn't applied to flights operated by OneWorld members or other partner airlines (because avios redemption table can be used for round trip routes only)?
Which strategy is the right?
a) Calculate the distance for the entire itinerary (sum of the distances of each segment) and determine the required number of avios;
b) Calculate the distance of each segment, determine the required number of avios for each segment and sum the number of avios of all segments.
