Price: well, you're paying, what can I say!
Sightseeing: You absolutely must go and see Akrotiri, unless you don't care about a ~3.5 K years old settlement. Other than that, ouzo, sunset, ouzo again, etc.

Also - is it possible to go to Sparta for a day trip? If so, how?
I would say not "worth" it per se: a very nice, deserted, medieval town on the mountai overlooking Sparta (Mystras) but Sparta as such has nothing to offer. Nice trip through the Peloponese.

...Plan:
May 7th: arrive in Athens at 1:25PM
Transfer to Hotel for 1 night and explore Athens (4hrs)
Acropolis Museum and Acropolis, not more than that in 4 hours.
May 8th: Depart from Athens at 5:15AM and arrive at Santorini at 6:00.
Spend all day exploring Santorini, see sunset, dinner
May 9th: Depart from Santorini at 8:25PM and arrive at 9:10PM in Athens,
Check into hotel
All of trhe above: OK.
May 10th: see all major sites in Athens
Another 3-4 museums? Old town? Piraeus? Lycabettus? Agora? Cape Sounion perhaps? Sort of pushy!
May 11th: Day trip to Sparta or Delphi
See above: I'd go to Delphi, much more impressive.

Needless to say, I leave Turkey for the Turks to answer.
