It depends on the airline that operates your flight from Darwin to Singapore. In many cases there are separate baggage drop counters for passengers who have already done check-in online or with machine and those typically have shorter and faster moving queues but I have no idea about Darwin airport.
But in general if you haven't flown much it's better to show up early. Check from your operating carrier about check-in times they recommend. It is much more relaxing to travel when you have time to spare. And after so many questions, please let us all know how your flights went.
