Best Way To Use My VS & AMEX Points?

I've tried to figure this out on my own, but I keep getting confused in the mass of restrictions/taxes/etc on VS.

I have 32,916 VS Flying Club Miles. (I could probably push this another 500 miles via e-rewards if needed)

I also have plenty of AMEX MR points (and theres a rumor about 20-30% Bonus to transfer to VS right now)

What would be the best way to use this?

I've read that buying a Premium Economy ticket, with a Upgrade to Upper Class is the best? What about Just buying the Economy -> Upgrade to Upper Class?

I know the Fuel Surcharges/Taxes are killer for PE/UC and that to upgrade you have to have a full Y ticket.

The route would be LAX-LHR.
