The upgrade should come within a few weeks of the issuance of your new card at the time the points are posted to your IHG account. One day you are Club, the next day Platinum. I believe bonus points posted will be determined by your status at check in or at the time the points are credited to your account, usually five days or so after check out.
Room upgrades will depend on issues such as availabilty and how well the individual property monitors the fact that upgrades are a perk of Platinum.
