Yes, while I've never done a mattress run I prefer to not have house keeping come in the room normally and I just put the do not disturb sign and have had no problems.
This might be getting a bit carried away, but the Hyatt Place I stay at delivers a morning newspaper to the room and typically only puts it half way under the door. Might look suspicious if a few days worth of papers are piled up. Maybe tell the front desk you don't want any when you check in?
Also, if you plan on doing this again at this hotel, maybe just talking to the manager and letting them know what your doing would be good. They're happy because they're getting revenue and an unoccupied room for a couple nights and your happy because you get the credit. I've read on other forums where people build a good rapport with a hotel and have been able to book a night or two and never show up and still get the credit. I think this maybe a rare occurrence, but it still may not hurt to introduce yourself and try to build a relationship.
