True but don't wait more than 20-30 min for such a bus. There are other options and there might be one going to a hotel right next door. Although some buses go to more than one hotel.

The NEX is not optimal for Shinjuku. The ride is about 90 min. And it's quite far from the train station to Park Hyatt so you are looking at a 2 hour plus door to door trip. What you could do is get off at Tokyo Station and take a cab. On the expressway it's fast and it dumps you off right by the hotel in Nishishinjuku. I drove from near Tokyo St. to Nishishinjuku a few weeks ago and it took me like 10 min around 4 pm.

The Keisei Skyliner is about 65 min assuming a quick connection but then you have to deal with the Yamanote commuter line with luggage for 20 minutes.
