I've been waitlisted many times for J class and have almost always been able to get the upgrade. They have contacted me by either phone, sms, or email to notify me to give them a call.

As for the upgrading immediately after a flight, I believe that TK will credit you the miles right away so that you can upgrade on the next leg. I have no experience with this, but recall seeing this info before.
