You can also upgrade at the airport. As I understand it, they will upgrade you if there are any seats at all available. I've done that on a IST-PVG flight. It was all done in the TK lounge at IST for me.
Regarding upgrade at airport/gate, flying JFK-IST-BKK (trying to upgrade IST-BKK), suppose I don't have enough miles at JFK, but do after completing JFK-IST flight, will gate agent count miles just completed from JFK-IST, or I have to wait until my miles to show up upon email notification? If it's the latter, it usually takes 3 - 12 hours after TK flight.
