Two main things I'd love:
(1) Basically Emma's idea: an easy user interface to a "show me where to go" feature. I tell the site my budget, dates, and which alliances I'm willing to fly...and it shows me real, available results. Seems doable, since it basically blends the old Travelocity Dream Map (which always showed low theoretical fares) and ITA queries that anyone can do with a little research.
(2) The next one - probably the tougher one - is a tool where I can provide vague requirements for a big award trip, provide my Award Wallet login (which has balances and elite statuses), and see some options. I want to go to any European gateway for a week and then on to Hong Kong for a week, 2 seats in J or F, anytime in a 3 month window. Willing to fly split up from my companion if flight times are similar enough. A heavily 'roided-up version of Awardnexus/KVS/Expertflyer/etc... Something the airlines probably wouldn't go for.
