If the route was
YVR (AC) --> LHR
LHR (LH) --> FRA
FRA (LH) --> JNB
could a travel book the AC leg in B (so I could instant upgrade if available) and the LH segments in U (lowest economy) or does the fare class have to be the same for all flights under a PNR?

Booking the TATL flights in B and the flights from Europe to SA would require two tickets AFAIK which could be in the same PNR if done by a very good TA. However, youŽd basically be paying for a TATL B fare and a U fare Europe-SA, having everything in one PNR would be of some advantage in case of irrops etc., though it doesnŽt sound like a particularly good/cheap option IMO.
IŽd recommend either of the following options:
1) purchase a H or M fare YVR-YYZ-ZRH(AC)-JNB(LX). The ZRH-JNB segments can be upgraded easily using miles if thereŽs I availability and a H/M should result in reasonable upgrade chances on the AC segments.
2) Depending on how much a H/M fare YVR-JNB costs, you should look into the *A YRTW fares which book into H or M as well.
3) purchase a B fare with the same routing if upgrading to J at time of booking for the AC flights is important to you
