I have checked in as early as 7AM at some properties (albeit airport ones) and most of the time my assigned room (upgraded) was already ready.

On non-airport properties where my assigned room wasn't ready I always have been offered the alternative of either waiting until it was ready or moving to another (upgraded) room.
