Qualified hotel stay

Fairly new to the hotel points/status game, trying to find out what constitutes a night's stay in terms of status level qualification.

1) if doing a mattress run for 1 night -- can you just check-in and then check-out, or do you have to stay overnight and check out the next day?

2) if you're don't show up and never check in (delayed flight, volunteer bump, etc.). Assume you did not or could not call and cancel the reservations, and your card will be charged for that night's stay.
