I would be surprised if they aren't able to hold a room until check-out time, if you've paid for the previous night and guaranteed it with a credit card. I'd continue to try until I got to talk to the Manager in person, then call around midnight of travel to remind them. You may have better luck with a larger/chain hotel than a small independent that may go through front desk clerks by the dozen.
