There's more than one route, but my suggestion is:
I280 east to
Garden State Parkway south to
I78 east to
NJ Turnpike south to
Goethals Bridge/Staten Island Expressway east to
Belt Parkway east to
JFK
(NB - when crossing Verrazano take upper level and stay in left lane for Belt Parkway)
This is a good route for that time and destination. Tolls would be $13 for the bridge and if there is anything on the GSP it'd be a $1 or $2. Beware that NYC traffic is unpredictable so listen to something like 1010AM for traffic updates.
