Wanted to add (what will be obvious to most) is that to make sure that using your card won't in effect jack up the price in some other way (meaning that they'll agree to take your card but won't give you a good deal for your trade in, etc).
What I did this time round, was to email several Internet Fleet mngrs, never mentioning the AMEX issue, getting their offers for selling price and getting an indication of what they would give me for my trade in. (And I would forward one dealer's offer to the next asking them to beat it. That was fun.) Then I took my car around to 3 places that were the most promising to get their offer for my trade in. THEN I brought up the AMEX issue. That way I was confident that using my card wouldn't "cost" me anything extra. The dealer that said he would only take $2500 on the card said that his store would take more on a credit card in OTHER situations, but that he couldn't take more AND give me the same good deal on the car and trade in. But like I said in my first post, there was a dealer that took $5000 and his out the door price did beat the others. (But sure would have liked to put more on the card. Be interested to hear how others were more successful so I can learn for next time.)
