I would absolutely suggest indicating that it's your anniversary. If you book via the Internet, there should be a blank for comments (or something to that effect). Add a sentence here about it being your anniversary. If you book over the phone, it never hurts to tell the reservationist to note the occasion in your booking. They will do this just like you were a guest asking for a room close to an elevator, on a lower floor, etc.
You also may want to register as a Starwood Preferred Guest before you book and travel.

You may not luck out and get the penthouse (and believe me this was luck), but from everything I've read and experienced about this hotel, I'll bet you get something.
