Going up Howe Sound via Squamish, Whistler and Pemberton takes you through the Coast Mountain range. Get into the Fraser Canyon at Lilloett & Cache Creek and on to Kamloops. This would be my favored route as I like the coastal range.
I'll second this route as the most scenic - Howe Sound, then moutains - Duffy Lake road is gorgeous:

While doable in a day, it is the longest route though in terms of time - Probably 7 hours driving time, 9 hours with some stops along the way.
