If there is upgrade inventory available in advance, you can use the PMU (I guess SWU now) to a CONFIRMED seat up front. This is where an EF subscription helps and it shows you which flights have availability. I just did this for flights on March 4 and March 10. Checked EF, booked on DL.com, called PSS and confirmed I was using PMUs. Turned in PMUs at airport today.

If there is NO upgrade inventory availablie, supposedly you can use them on the day of departure at the airport if there are still seats available up from (this is something new for 2010).
