Not sure what would happen if you call them and release the seat and then call right away and put it on hold again.

Each airline has its own hold policy, so if you do this, be sure you don't exceed one of the hold policies for the airlines in your itinerary.

SQ, for example, does not allow holds whatsoever. If you get an agent to hold a SQ flight for you, it may be gone when you call back.
