In my experience, no, they won't have the car pre-setup for you.
At Hertz MCO in particular, I had to hunt down an employee who took me to a room with a pile of carseats. I grabbed one and installed it myself.
I quickly learned to adjust my overall carseat approach: we now travel with our own (two of them), as inconvenient as I used to think that was.
This is not unique to Hertz: I see the "big pile of carseats" at National and others as well. I get the sense that affixing seats to actual cars prior to the renter showing up would cause too many problems for existing systems.
