Whole process was pretty straightfwd for me and my fam. Did the application online, got the conditional approval within 2 days, made appointments at YVR, which was about 2 week wait for time slots of 4.
Could have done it at the border, without waiting the 2 weeks. But would have to stop in at YVR to do retina scan. Did the interview, which is a formality, review docs and that's it. Got cards within a week of the interview.

*Applied on-line Dec 4th 2009, approved Dec 10, interviewed Dec 10 (got lucky and was flying out at that time ). Now just gotta wait for the card to be mailed to me, which the agents thought would take forever.
Having shared that... I'd suggest that with the recent issues, it would be very unlikely that you could get your NEXUS within the next 16 days. But... if you have future travelling planned, why not give it a try?
