You get only one free transfer. And a paper transfer is only valid from bus to bus; only Metrocard gives a free transfer between bus and subway. If the Q33 and Q53 fareboxes are both working, then the trip via the Q33, the Q53, and the A train would cost two fares, or $4.50, rather than $2.25.
Depending on the day of week and time of day, the Q33 to Q53 to A train route would probably save about 30 minutes over the routing via Manhattan.
