Ever since I had to drag a suitcase uphill in the rain because the shuttle advertised on the hotel's website only ran on weekdays, if I think I'm going to need it, I always call in advance and ask about the days/hours it operates, whether they take advance reservations, and, in the case of airport shuttles, whether it's on a fixed schedule or on demand.
