I think that you are eligible for Diamond amenities (if a Diamond), i.e. upgrade to RC if available, but that you cannot combine the gift certificates with any other certificate, i.e. with a suite upgrade coupon sent to you as Platinum. I haven't seen anything in print that gives you the ability to either upgrade for $$$ either on a confirmed basis or on check-in, so I don't think it is available, at least officially.
