Well, at least the US outstations (most recent experience @ JFK) are supposed to check the fare rules now and look for the respective endorsement.
If the fare rules of your Z fare state that it's not upgradeable, they won't let you use SWUs, mileage or even LH e-vouchers on the day of departure (the LH upgrade instruments get blacked out in the MAM Direct upgrade mask).
So if you're travelling on a Z fare ex USA that has the "non-upgradeable" provision, expect to be seated in Business Class except for an oversale situation... (during which they might gladly take your SWU rather than hand out a free op-up)
