Unless Die Bahn has changed in the three years since I did the exact same trip, you take one train from Frankfurt to Nuremberg. There, you change to a train directly to Prague.
You can do this trip a little cheaper on weekends by buying the "Happy Weekender" ticket. That would eliminate the need for a separate fare from Frankfurt to Kahl, as it is good for travel anywhere in Germany.
