My Diamond status kicked in last Wednesday (5/13). I had 2 stays this past weekend, and both voluntarily mentioned my Diamond Status at check-in and gave me the amenities as indicated in my profile without me making any modifications to either record.

Thus in short, you should expect to see the benefits automatically kick in the day or day after your status updates.
