Are you really sure you want to stay in Rothenburg for two nights? The town is tiny and you can see everything of interest in a day (don't miss the Medieval Criminal Museum to have a look at torture instruments from days gone by!). If you take an early morning train from Frankfurt you should be there before noon so even with one night you could enjoy a whole evening and a morning without the tourist masses. This way you could fit in a night in e.g. Würzburg which is just in the middle between Frankfurt and Rothenburg and a beautiful city. Just my opinion of course.

For beer in Rothenburg: I can't recommend any specific place, I think the ones in the historic centre are probably all nice and traditional (and they should all serve some Franconian wurst with sauerkraut). Try to get your hands on some "Aischgründer" beer, it's delicious!
