Yes and no. There are a lot of contributing factors to turbulence and potholes.

Flying into DEN, for example, is almost always bumpy because of the terrain and winds around there.

Just like roads in Chicago or Detroit are more likely to have potholes than say LA because of the snow, ice, and road treatments they use there.

I don't think the OP's question is that outlandish. There are probably routes that are typically calm due to weather patterns and other factors. Obviously it's not exact, but he's going after the typical or most likely.
