If by ticket counter you mean check in, yes. Because that is where the routing is attached to the bag.
In theory, the gate agent could send a "message" to the bags department. In practice, it still goes on the routing of the bag. That means also, because you switched your flight, they won't deliver the bag to you.
Trust me -- stick to the original routing.
