If you fly on a single ticket your luggage will be checked through to TXL. still telling us whcih airlines would help to answer your question more specific. For example if you fly with Lufthansa you can also ask your question here: Is this [FRA] connection easy? [The FRA LH connection Q&A thread]

So in FRA it will be just immigration. Customs will be done in TXL (if at all, unlike in the US you don't have to fill out a customs form. You just walk through the "nothing to declare" lane. Of course only if you have nothing to declare.)

Also flights from the US tend to arrive earlier especially in fall and winter due to stronger winds over the North Atlantic. So there is a good chance you will arrive 30 minutes earlier.
