One other comment regarding lines - you can get the Roma Pass at many locations in the city, and use it directly at the turnstiles at the Colosseo/Palatino archaelogical sites. When you get to these sites, just take the Roma pass to the turnstile and scan the barcode - we had practically no wait despite near hour-long lines for the ticket windows.

It is only 20 euro, and includes full use of public transport.
Agreed about getting the Roma Pass. But in case there is any confusion, you cannot use it at the Vatican Museum. Different country, you know.
