Largest I can remember that wasn't hub to hub...
Had 61 on the UG list ORD-CLE last week on a B737.
As a Gold with an RPU that had been waitlisted since October- I was 1st on the list of with 4 seats from T-48 down to T-2 when I got shifted down to 2nd on the list and 3 others cleared. I can only guess some GSes SDCed?

complete outlier here. This one made absolutely no sense whatsoever. 86 on the upgrade list MSP-ORD on a Wednesday 3pm flight. This was on a 738. Two GS on the flight, (me and another guy) Group 1 was a mile long. We both commented that we had never seen it like that on a Wednesday. Sure a Friday or Thursday, but a midweek middle of the day flight.

The displayed gate list only shows those that have checked in (or for CPU flights, also those cleared). A late check-in would have create the same effect. As would a late purchase.

72 on SFO-DEN at 12:55 p.m. on a Monday, 1/23. It was a 777 with lie-flat seats.
We were on Global First Saver awards and ended up sitting in Coach because this segment never cleared. The agent at the Global First Lounge said there were a lot of Saver First passengers on the First Standby list, which is supposed to be cleared ahead of the upgraders. I didn't see any Saver First awards on any flight on 1/23, even though I checked weekly from November onward.

I have often seen SJC-DEN at 70 people on a Thursday on an A319/320(50% of all pax)..
The longest standby list I have seen was 250+ people for ORD-DEN on a sold out 2 cabin 777 in 2009, it was middle of July after thunder storms. Don't even remember what they were offering for VDBs.

I had 123 on a DEN-EWR 777 a few weeks ago. I was on a V fare as a 1K and was #19 when the door closed (admittedly I had SDC'ed to this flight during the day to get home earlier, so I was just happy to snag E+ as we went out full). At least 10-12 names at the bottom were NRSA and I noticed lots of commuting crewmembers.
10-11 pax appeared on the cleared list out of 28 seats in the cabin. DEN is usually a cheap P fare.

01/23- UA 550, 737-900, the gate agent announced that there were 127 passengers in group 1 and 2 and I remember there were ~110 passengers on upgrade list.
I was #27 on this list on an S fare (1k)

Best I've seen was ~150 people on the upgrade list. It was SFO->HNL on the Saturday morning of President's Day weekend this year. First class on the Hawaii-configured 777 was full but Economy was less than half full until over 100 non-revs were cleared from the standby list. Since they were all on the upgrade list as well, it was pretty massive.

137 on UA 408 this morning between SFO and IAD. Plane swapped for the new lay flat Hawaii config. I am 14 on the list as a 1k.
