I have had memorable (as in, memorably good) experiences in New York, Chicago, San Diego, Toronto and Mexico City. And am about to go to SF with my GF, which should be rather good!
To pick one..? Hmm, not sure I could.

Places I've liked and would return to in a heartbeat:
Boston
Chicago
NYC
Seattle
Toronto
The Twin Cities
I also like and highly recommend my hometown, Milwaukee (except maybe not in Dec, Jan, Feb or March if you're not a cold weather/snow person) and my current home, Washington, DC (except maybe not June - August if you don't enjoy heat and horrendous humidity).

Toronto! Hands down the best. I want to end my days there.
New York city, because it reminds me of Toronto.
Baltimore, for architecture, bars and food.
Las Vegas, it is one of those "must see before you die"

Buffalo is in my top 5 most LIKED actually.
Great food, arts scene and low cost of living and close to huge parks and The Falls. Sure Winter is cold, but then again, so if 3/4 of the USA in the winter...
TOP 5
NYC--Its unique in so many ways
San Diego--Urban downtown, great beaches
Chicago--Almost NYC but nicer people
SF--Awesome views, romantic etc
Buffalo--My wild card. Has its charm--just need to know where to go.

Pick up a car and spend all your time in CA. I know, CA is not a city. Take Highway 1 and do San Francisco to San Diego. In between, stop by Monterey/Carmel, Santa Barbara, LA (I mean Santa Monica, Venice, Malibu, Laguna Beach) and of course San Diego.
