Yes it’s valid. I flew it about 3 weeks ago in Suites with my wife on our honeymoon. I booked JFK - FRA - SIN the day award space opened and tacked on SIN - SYD as a stopover addition. SIN - SYD was on SQ231 (the late night departure).
Of course that booking was 11 months ago so current award availability may have changed but routing is definitely valid.

Since SQ applies married segment logic to award inventory (which is a huge PITA), you may want to check the following options as well.
JFK-FRA (>24h stopover) - SIN - SYD
JFK(-FRA)-SIN (>24h stopover) - SYD
JFK-FRA (>24h stopover) - SIN - SYD
JFK(-FRA)-SIN (>24h stopover) - SYD
